Product Specific Information
The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimer or CD8αα homodimer.
CD8 interacts with MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T cell development and activation.
Applications Reported:
Used for flow cytometric analysis and immunohistology staining of frozen tissue sections.
Applications Tested:
Validated by flow cytometric analysis of mouse splenocytes at ≤0.125 µg/test (100 µL final volume).
Optimal performance requires titration based on cell number (10⁵–10⁸ cells/test).
Fluorophore Information:
eFluor® 660 replaces Alexa Fluor® 647, emits at 659 nm, excited by 633 nm red laser.
Excitation: 633–647 nm; Emission: 668 nm; Laser: Red Laser.
Post-manufacturing filtration: 0.2 µm.
Target Information
CD8 (Cluster of differentiation 8) is a type I transmembrane glycoprotein involved in T cell differentiation and activation.
It acts as a co-receptor with TCR for MHC class I molecules and is expressed on T cells, NK cells, monocytes, and dendritic cells.
CD8 ligation triggers Lck recruitment, leading to lymphokine production and cytotoxic T lymphocyte activation.
CTLs are critical for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells is enhanced by IL-2, IL-12, and TGF-β1.
