Product Specific Information
The monoclonal antibody reacts with human AITR (Activation Inducible TNFR family member), a ~25 kDa member of the TNFR superfamily. AITR mRNA is detected in lymph node, peripheral blood leukocytes, and weakly in spleen. Expression is upregulated within 24 hours after PMA/ionomycin or PHA stimulation of PBMCs. At the protein level, AITR is expressed by a small population of activated PBMCs. AITR associates with TRAF1, TRAF2, and TRAF3, inducing NF-κB activation via TRAF2. TL6 (AITRL) has been reported as its ligand, mediating cross-talk between T lymphocytes and endothelial cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry using stimulated normal human peripheral blood cells. Use 5 µL (1.0 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test (empirically determined).
Light Sensitivity: Protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark. Clone-specific performance should be confirmed empirically.

Target Information
Tumor necrosis factor receptor superfamily member 18 (TNFRSF18), also known as GITR or AITR, is encoded by the TNFRSF18 gene located at 1p36.33. This receptor shows increased expression upon T-cell activation and plays a key role in immunological self-tolerance maintained by CD25⁺CD4⁺ regulatory T cells. Knockout studies in mice indicate its role in regulating CD3-driven T-cell activation and programmed cell death. Three alternatively spliced transcript variants encoding distinct isoforms have been reported.
