Product Specific Information
The II/41 monoclonal antibody reacts with the µ heavy chain of mouse IgM. It does not react with other classes of mouse immunoglobulin including IgD, IgG, or IgA. IgM is expressed intracellularly during early stages of B lymphopoiesis and on the surface of mature B cells in bone marrow and peripheral tissues.
Fluorochrome-conjugated II/41 can be used as a detection secondary for mouse IgM.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse spleen cells. Recommended ≤0.125 µg/test (100 µL final volume). Optimize empirically for best performance.
Light Sensitivity: Tandem dye is photo-sensitive; protect vial and samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
Excitation/Emission: 488–561 nm / 775 nm (Blue, Green, Yellow-Green lasers).
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Mouse IgM is an immunoglobulin M antibody produced by B cells in response to antigens. It is the first antibody produced during an immune response and is the largest immunoglobulin in serum. Composed of five heavy and five light chains, it plays a key role in complement activation and antigen agglutination.
Widely used in research and diagnostics (ELISA, Western blotting, immunohistochemistry).
