Product Specific Information

The monoclonal antibody 3D10 recognizes Ly-49H, a member of the C-type lectin Ly-49 multigene family of receptors found on natural killer (NK) cells.
Ly-49H, like Ly-49D, lacks the ITIM motif and contains an arginine in the transmembrane domain, indicating its role as an activation receptor.
It is expressed in a subset of NK cells but not NKT (NK1.1/CD3+) cells. Expression confirmed in C57BL/6 and NWNA mice, not in Balb/c or DBA/2 strains.
No cross-reactivity to Ly-49A, C, D, or G2 observed. Addition of 3D10 induces target cell lysis.

Applications Reported: Flow cytometric analysis.
Applications Tested: Mouse splenocytes; ≤0.5 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Light Sensitivity: Tandem dye is photo-sensitive. Protect vial and samples from light.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54).

Target Information

Ly49H is a C-type lectin receptor of the Ly-49 family found on NK cells.
Functions as an activation receptor due to absence of ITIM and presence of transmembrane arginine.
Expressed in NK cell subsets, not in NKT cells (NK1.1/CD3+). NK cells may express multiple Ly49 proteins.
