Product Specific Information

  • For FACS analysis, use 10 µL of the suggested working dilution to label 1×10⁶ cells in 100 µL.
  • Rat anti-Human CD8 antibody, clone YTC182.20 recognizes the human CD8 cell surface antigen (~30/32 kDa glycoprotein) expressed by the cytotoxic/suppressor subset of T lymphocytes, and more weakly by NK cells.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein belonging to the immunoglobulin family of receptors. It plays a critical role in signal transduction, T cell differentiation, and activation.

  • CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with T cell receptor (TCR) for MHC-I molecules.
  • CD4, in contrast, serves as a co-receptor for MHC-II molecules.
  • CD8 can also exist as disulfide-linked homodimers of CD8α on subsets of T cells, NK cells, monocytes, and dendritic cells.
  • Engagement of MHC-I/peptide complexes by CD8 triggers recruitment of lymphocyte-specific protein tyrosine kinase (Lck), leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
  • Activated CTLs are essential for pathogen and tumor cell clearance, with differentiation of naive CD8⁺ T cells enhanced by IL-2, IL-12, and TGF-β1.
