• Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Beta-2-microglobulin (B2M) is a 12–14 kDa protein that forms part of the MHC class I complex. It is expressed on various cell types including lymphocytes, thymocytes, monocytes, granulocytes, platelets, endothelial, and epithelial cells, but absent on erythrocytes.
It is also found in free form in plasma and shares homology with immunoglobulin constant regions, forming a predominantly beta-pleated sheet structure.
B2M is involved in antigen processing and presentation, TCR signaling, and downstream signaling in naive CD8+ cells. The B2M gene is located on chromosome 15, and mutations can lead to hypercatabolic hypoproteinemia.
Beta-2-microglobulin levels are useful for assessing renal function and as a prognostic marker in HIV infection.
