Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Beta-2-microglobulin (B2M) is a 12–14 kDa protein that forms part of the MHC class I complex. It is expressed on various cell types including lymphocytes, thymocytes, monocytes, granulocytes, platelets, endothelial, and epithelial cells, but not on erythrocytes. B2M is also found in plasma in free form.
Its amino acid sequence is homologous to immunoglobulin constant regions, featuring a predominantly beta-pleated sheet structure. B2M participates in antigen processing and presentation, TCR signaling, and downstream signaling in naive CD8+ cells.
The B2M gene is located on chromosome 15, and mutations can cause hypercatabolic hypoproteinemia. B2M levels are used as markers of renal function and as prognostic indicators in HIV infection.
