Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
Suggested positive control: bone marrow or lymphoma.
Target Information
Spleen tyrosine kinase (SYK) is a cytoplasmic tyrosine kinase, along with ZAP70, considered a potential drug target for allergic and autoimmune diseases. SYK may also play a role in cancer.
Upon B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI) activation, SYK translocates to the plasma membrane and phosphorylates downstream adaptor proteins, initiating signaling pathways such as calcium mobilization, cytoskeleton remodeling, and gene transcription.
SYK interacts with receptor ITAM motifs via its SH2 domains, which are phosphorylated by Src-family kinases that also activate SYK through phosphorylation of its activation loop.
