Heat-mediated antigen retrieval is recommended prior to staining using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with the primary antibody for 10 minutes at room temperature.
A suggested positive control is breast carcinoma.

Target Information

STAT5 alpha is a member of the STAT family of transcription factors. In response to cytokines and growth factors, STAT family members are phosphorylated by receptor-associated kinases and then form homo- or heterodimers that translocate to the cell nucleus, where they act as transcription activators.
STAT5 alpha mediates responses to ligands such as IL2, IL3, IL7, GM-CSF, erythropoietin, thrombopoietin, and various growth hormones. Activation of STAT5 alpha in myeloma and lymphoma associated with a TEL/JAK2 gene fusion occurs independently of cell stimulus and is essential for tumorigenesis.
The mouse counterpart of STAT5 alpha induces expression of BCL2L1/BCL-X(L), suggesting an antiapoptotic function. STAT5 alpha and STAT5 beta show 96% sequence similarity and are co-expressed in most tissues, though differential accumulation has been observed in muscle and mammary tissue.
STAT5 alpha plays critical roles in reproduction, lactation, immune function, and somatic growth.
