Product Specific Information
The DX5 monoclonal antibody reacts with CD49b, an antigen expressed on a majority of mouse natural killer (NK) cells and a subset of T cells. DX5 reacts with all strains of mouse tested, including BALB/c, C57BL/6, C3H, CBA, DBA, AKR, SJL, and 129.
Simultaneous staining of C57BL/6 spleen cells with anti-NK1.1 mAb (PK136) and DX5 reveals coexpression of both markers by most cells, as well as small populations of DX5⁺PK136⁻ and DX5⁻PK136⁺ cells.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤1 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.
Fluorochrome Information:
eFluor™ 506 is excited with a 405 nm violet laser and emits at 506 nm. Recommended filter: 510/20 band pass or equivalent. Ensure instrument compatibility.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD49b (Integrin alpha 2) is a glycoprotein (~150 kD) forming a heterodimer with CD29 (Integrin beta 1) to create VLA-2 (Integrin α2β1). This complex acts as an extracellular receptor for laminin, collagen, and fibronectin, mediating adhesion and intracellular signaling.
CD49b plays roles in VEGF-induced angiogenesis, adhesion, and lymphocyte activation. It is expressed by NK cells, NK-T cells, monocytes, platelets, epithelial cells, and subsets of CD4⁺ T cells, including Tr1 regulatory cells co-expressing LAG-3.
