Product Specific Information

The YKIX302.9 monoclonal antibody reacts with canine CD4, a cell surface receptor expressed on thymocytes, mature T cells, dendritic cells, and neutrophils. CD4 binds to MHC class II molecules and plays an essential role in T cell development and homeostasis.

This antibody has been reported to block MHC II-restricted T cell responses both in vivo and in vitro.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry using normal canine peripheral blood cells at 5 µL (0.06 µg)/test.
A test is defined as staining a sample in 100 µL final volume. Cell number may vary from 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended filter: 660/20 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in dark with minimal impact on brightness and FRET efficiency.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. It is mainly expressed on T helper cells but also on monocytes, macrophages, and dendritic cells. At the tissue level, CD4 is found in thymus, lymph nodes, tonsils, spleen, and certain regions of the brain and gut. It plays a key role in initiating T-cell activation and may mediate neuronal damage in immune-related CNS diseases.
