Description:
This DS5MMER monoclonal antibody recognizes mouse MerTK, a 170–210 kDa member of the TAM family of tyrosine kinase receptors that includes Axl and Tyro3. MerTK is expressed on tissue macrophages and is involved in the removal of apoptotic cells via ligands Protein S and Gas6. Upon ligand binding, MerTK undergoes autophosphorylation activating PI3K and Akt pathways, leading to phagocytosis and inhibition of TLR-induced cytokine production. MerTK deficiency leads to autoimmunity and inflammation. It is also expressed on malignant cells and may play a role in immune evasion.

Applications Reported:
This DS5MMER antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of resident peritoneal macrophages. Recommended ≤0.5 µg/test (100 µL final volume). Cell number can range from 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited with the 405 nm violet laser and emits at 645 nm. Use a 660/20 bandpass filter. Protect from light due to photo-oxidation sensitivity.

Recommended Buffers:
When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

MERTK (cMER) is a tyrosine kinase proto-oncogene involved in retinal pigment epithelium (RPE) phagocytosis and immune regulation. It regulates cell survival, migration, differentiation, and efferocytosis. Ligand binding induces autophosphorylation and downstream signaling via GRB2 or PLCG2, activating MAPK, FAK/PTK2, and RAC1. Plays roles in macrophage clearance, platelet aggregation, and suppression of TLR-mediated immune responses through STAT1 activation and SOCS1/3 induction.
