Product Specific Information

The LN3 monoclonal antibody reacts with human MHC class II molecule HLA-DR.
HLA-DR is expressed on antigen-presenting cells (APCs) such as B cells, monocytes, macrophages, dendritic cells, and activated T cells. It is a heterodimeric transmembrane protein composed of alpha and beta subunits, playing a key role in peptide presentation to CD4⁺ T lymphocytes.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells
    • Recommended usage: 5 µL (0.125 µg)/test
    • A test = staining of cell sample in 100 µL final volume (10⁵–10⁸ cells/test)

Super Bright 645 Characteristics:

  • Tandem dye excited by violet laser (405 nm), emission at 645 nm
  • Recommended filter: 660/20 bandpass
  • Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes
  • Light-sensitive: protect from light
  • Fixation: compatible with IC Fixation Buffer (#00-8222) or Fix/Lyse Solution (#00-5333) for up to 3 days at 4°C

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).
The alpha chain gene has 5 exons encoding leader peptide, extracellular domains, transmembrane domain, and cytoplasmic tail.
DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5, while polymorphisms reside in the beta chain defining peptide binding specificity.
Hundreds of DRB1 alleles are known and routinely typed for transplantation compatibility.
HLA-DR is primarily expressed on APCs such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
MHC class II molecules (DR, DQ, DP) present processed peptides to T-helper cells and are essential for initiating immune responses.

HLA and MHC antibodies are crucial in Immunopeptidomics, enabling identification of neoantigens via HPLC-MS/MS.
