Product Specific Information
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease regulation in mice. It is mainly expressed on activated T and B lymphocytes.
PD-1 ligands include PD-L1 (B7-H1) and PD-L2 (B7-DC). J43 inhibits the binding of PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells. In vivo, J43 enhances contact hypersensitivity and acute GVHD, and can exacerbate autoimmune models such as EAE and NOD diabetes.

Handling and Storage Notes:

Target Information
T-cell activation is regulated by inhibitory receptors such as PD-1 (PDCD-1), which binds to ligands PD-L1 and PD-L2 on antigen-presenting cells. This interaction suppresses immune responses in the absence of danger signals. PD-1 deficiency leads to hyperactivated immune systems and autoimmune diseases. Though predicted at 55 kDa, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
