Product Specific Information
The MIH4 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the immunoglobulin superfamily. PD-1 contains ITIM and plays a key role in peripheral tolerance and autoimmune disease. It is expressed predominantly on activated T and B lymphocytes. PD-L1 (B7-H1) and PD-L2 (B7-DC) are known ligands of PD-1.
This MIH4 antibody is optimized for flow cytometric analysis and has been pre-diluted for use at 5 µL (1.0 µg) per test. One test corresponds to staining a cell sample in a final volume of 100 µL, typically 10⁵–10⁸ cells/test.
Super Bright 645 is a tandem dye excited with a 405 nm violet laser and emits at 645 nm. Use a 660/20 bandpass filter for detection. When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
- Light Sensitivity: Protect vial and stained samples from light.
- Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness.

Target Information
T-cell activation is regulated by PD-1 (PDCD-1), an inhibitory receptor expressed on activated T cells. PD-1 binds ligands PD-L1 and PD-L2 on antigen-presenting cells, suppressing immune responses in the absence of danger signals. PD-1-deficient mice exhibit hyperactivated immune systems and autoimmune disease. Despite its predicted molecular weight, PD-1 often migrates at higher molecular weight in SDS-PAGE.
