Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
The DAXX gene encodes a multifunctional protein localized in both the nucleus and cytoplasm. It interacts with proteins such as apoptosis antigen Fas, centromere protein C, and transcription factor E26 oncogene homolog 1. In the nucleus, DAXX acts as a transcriptional repressor that binds to sumoylated transcription factors. Its repression can be relieved by sequestration into promyelocytic leukemia nuclear bodies or nucleoli. DAXX associates with centromeres during G2 phase and may regulate apoptosis in the cytoplasm. Its localization and function are modulated by post-translational modifications including sumoylation, phosphorylation, and polyubiquitination. Multiple transcript variants exist due to alternative splicing.
