Description:
The monoclonal antibody eBio440c recognizes Siglec-H, a protein exclusively found on plasmacytoid dendritic cells (pDC) or type I IFN-producing cells (IPC) in the naive mouse. Mouse IPC are typically PDCA+, CD11c+, CD11b-, B220+, and Ly-6C+, and respond rapidly to viral infection.
Siglec-H is a transmembrane protein of the Ig superfamily that, like CD33, binds sialic acid but lacks the cytoplasmic ITIM domain. To enable signal transduction, Siglec-H associates with DAP12. The eBio440c antibody has been shown to inhibit pDC function, reducing IFNα secretion in response to CpG stimulation.
Some mouse strains (e.g., SJL) exhibit higher percentages of pDCs compared to C57BL/6.

• Flow cytometric analysis.

Applications Tested:

• Blocking staining of the direct conjugate of eBio440c.
• Use ≤ 2.0 µg/test (100 µL final volume).
• Cell number: 10⁵–10⁸ cells/test (empirically determined).
• Optimize antibody titration for best performance.
• Direct conjugates such as Product #11-0333-82 are recommended for flow cytometry.

• Use in a sterile environment.
• 0.2 µm post-manufacturing filtered.
• Purity: >90% (SDS-PAGE).

Target Information

Siglec-H is expressed exclusively on pDCs or IPCs in naive mice. These cells (PDCA+, CD11c+, CD11b-, B220+, Ly-6C+) are key responders to viral infection.
Siglec-H, a member of the Ig superfamily, binds sialic acid but lacks the ITIM domain. It signals via DAP12 association.
The eBio440c antibody inhibits pDC function by reducing IFNα secretion upon CpG stimulation.
Strain differences exist, with SJL mice showing higher pDC levels than C57BL/6.
