Target Information
GRP78 is a 78 kDa glucose-regulated protein belonging to the HSP70 family. It resides in the endoplasmic reticulum (ER) and transiently associates with newly synthesized secretory and membrane proteins. GRP78 also interacts with mutant or misfolded proteins, marking them for degradation.
The conserved Lys-Asp-Glu-Leu (KDEL) sequence at the C-terminus ensures ER protein retention via the KDEL receptor. GRP78 plays a key role in protein folding, degradation of misfolded proteins, and maintenance of cell homeostasis, preventing apoptosis.
Reduced GRP78 levels are observed in Alzheimer’s Disease brains and are linked to PS1 gene mutations. Induction of GRP78 is associated with drug resistance to antitumor agents.
