Target Information
Sclerostin은 주로 골세포(osteocyte)에서 분비되는 당단백질로, Cerberus/DAN 계열의 BMP 길항제입니다. 이는 Wnt 신호전달 경로의 내인성 조절자로 작용하며, 골 항상성의 억제 조절자 역할을 합니다.
Sclerostin은 뼈, 골수, 연골, 신장, 간 등에서도 발견되며, 비대성 연골세포 및 cementocyte에서도 생성됩니다.
LRP5 및 LRP6과 결합하여 Wnt/β-catenin 신호를 억제하며, LRP4와의 상호작용을 통해 Wnt 신호 억제를 촉진합니다. 또한 RANKL 발현을 증가시켜 파골세포 활성을 촉진할 수 있습니다.
Sclerostin의 기능 이상은 sclerosteosis 및 van Buchem disease (VBD)와 같은 골형성 과다 질환을 유발할 수 있습니다.
