Product Specific Information

This product is preservative-free. It is recommended to add sodium azide (final concentration 0.05%–0.1%) to prevent contamination.

Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. Antibody DNA sequences from immunoreactive rabbits are cloned and screened to select optimal clones for production.
Advantages include:

  • Superior specificity and sensitivity
  • Lot-to-lot consistency
  • Animal origin-free formulation
  • Broader immunoreactivity due to rabbit immune repertoire

This antibody specifically recognizes Rat Sclerostin/SOST.

Target Information

Sclerostin is a glycoprotein mainly secreted by osteocytes and a member of the Cerberus/DAN family of BMP antagonists. It acts as an endogenous regulator of the canonical Wnt signaling pathway and bone homeostasis.

Although primarily expressed in osteocytes, sclerostin is also found in bone, bone marrow, cartilage, kidney, and liver, and can be produced by hypertrophic chondrocytes and cementocytes.
Sclerostin regulates the Wnt/β-catenin signaling pathway by forming inhibitory complexes with LRP5 and LRP6, essential co-receptors for Wnt ligands. It also interacts with LRP4 to inhibit Wnt signaling and promotes osteoclast activity by increasing osteocyte expression of RANKL.

Mutations causing reduced sclerostin levels lead to bone dysplasia disorders such as sclerosteosis and van Buchem disease (VBD), characterized by excessive bone growth and hypermineralization.
