Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a crucial role in the cellular response to genotoxic stress and is a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits CDK1 and CDK2, blocking the transition from G1 to S phase or from G2 to mitosis after DNA damage, allowing DNA repair. In the cytoplasm, p21 has an anti-apoptotic effect by binding and inhibiting caspase 3, ASK1, and JNK. p21 exhibits dual functionality in carcinogenesis, acting both as a tumor suppressor and an oncogene depending on context.
