Specificity Comments:
This monoclonal antibody recognizes a 21 kDa protein identified as the p21WAF1 tumor suppressor protein. It is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 acts as a specific inhibitor of CDKs and is involved in the pathogenesis of various malignancies. Expression of p21 inhibits the cell cycle during the G1 phase and is regulated by the tumor suppressor protein p53. Expression is induced by wild-type p53 but not mutant forms. Normal cells generally show intense nuclear p21 expression, while loss of p21 expression has been reported in many carcinomas (gastric, non-small cell lung, thyroid).
Target Information
Protein p21 is a tumor suppressor that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a key role in the cellular response to genotoxic stress and is a major transcriptional target of p53. Nuclear p21 binds and inhibits CDK1 and CDK2, blocking the transition from G1 to S phase or from G2 to mitosis after DNA damage, allowing DNA repair. In the cytoplasm, p21 exhibits anti-apoptotic effects by binding and inhibiting caspase 3, ASK1, and JNK. p21 has dual functions in carcinogenesis, acting as both a tumor suppressor and an oncogene depending on context.
