Target Information
Trpc6 forms a receptor-activated calcium channel in the cell membrane. The Trpc6 channel is activated by diacylglycerol (DAG) and is thought to be regulated by a phosphatidylinositol second messenger system. Activation occurs independently of protein kinase C and is not triggered by low intracellular calcium levels.
Defects in the Trpc6 gene are associated with focal segmental glomerulosclerosis 2 (FSGS2).
Trpc6 belongs to the mammalian transient receptor potential (TRP) superfamily, specifically the canonical TRP (TRPC) family. Members of this family are activated through PLC-coupled receptors and act as receptor-regulated cation channels.
The TRPC3/6/7 subfamily can be activated by DAG analogs, suggesting a mechanism of activation by PLC-coupled receptors. When expressed in transfected cells, Trpc6 functions as a non-selective, store-independent, receptor-activated cation channel, activated by DAG in a PKC-independent manner and insensitive to IP3 activation.
Increasing evidence suggests that TRPC6 encodes endogenous DAG-activated receptor-operated cation channels in vivo. Diseases associated with TRPC6 include Glomerulosclerosis, Focal Segmental, 2 and Focal Segmental Glomerulosclerosis.
