Product Specific Information

This NGZB monoclonal antibody reacts with mouse Granzyme B, a member of the granzyme serine protease family found in cytotoxic T cells and NK cells. Granzyme B is also known as CGL1 and CTLA-1, expressed after activation in cytotoxic T lymphocytes. It plays a critical role in apoptosis induction via mannose 6-phosphate receptor-mediated pathways.

Applications Reported: Intracellular staining and flow cytometric analysis.
Applications Tested: Tested with mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) following BestProtocols® Protocol A: Two-step protocol for cytoplasmic intracellular proteins.

Use ≤0.125 µg per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize antibody titration for best results.

Light Sensitivity: Protect from light; tandem dye is photo-sensitive.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.
Excitation: 488–561 nm

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Granzyme B is a serine protease found in cytotoxic T cells and NK cells. It induces apoptosis by cleaving caspase-3 and initiating the caspase cascade or via mitochondrial pathways through Bid cleavage. It is stored in specialized lytic granules and interacts with mannose 6-phosphate receptor functioning as a death receptor during cytotoxic T cell-induced apoptosis.
