Product Specific Information

This KIKSI0803 monoclonal antibody recognizes STAT1 phosphorylated on tyrosine 701. STAT proteins are activated by ligand binding to cytokine receptors associated with JAK kinases. Phosphorylation at Tyr701 by JAK1/JAK2 is essential for STAT1 dimerization, nuclear translocation, and DNA binding.

  • Specificity Determination: ELISA, flow cytometry, and western blotting
  • Reported Applications: Intracellular staining followed by flow cytometric analysis
  • Tested Protocols: Tested by intracellular staining using Protocol C (Fixation/Methanol). Protocol A and B are not compatible.
  • Recommended Use: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
  • Light Sensitivity: Protect from light due to photo-induced oxidation sensitivity.
  • Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57).
  • Excitation/Emission: 488–561 nm / 775 nm (Blue, Green, Yellow-Green lasers).

Target Information

STAT1 (Signal Transducer and Activator of Transcription 1) is a transcription factor activated by interferons and growth factors. It regulates genes involved in cell growth, apoptosis, immune response, and lipid metabolism. Located on chromosome 2, STAT1 activation occurs via phosphorylation by JAK kinases, enabling nuclear translocation and gene transcription regulation.
STAT1 responds to ligands including IFN-α, IFN-β, IFN-γ, IL-10, IL-6, IL-11, LIF, CNTF, G-CSF, EGF, PDGF, and CSF-1. Two alternatively spliced transcript variants encode distinct isoforms.
