Product Specific Information

The UT41 monoclonal antibody was generated against BaF3 cells overexpressing mouse TLR4. The Toll family includes at least twelve members, characterized by leucine-rich extracellular domains and cytoplasmic domains homologous to the type I IL-1 receptor. TLR2 and TLR4 act as pattern recognition receptors involved in innate immunity and inflammation.

TLR4 associates with MD-2 and CD14 to form a complex responsible for LPS recognition and signaling. TLR4 (~100 kDa) is expressed by myeloid lineage cells. The MTS510 monoclonal antibody co-immunoprecipitates MD-2 (~30 kDa) and TLR4 (~100 kDa) and reacts preferentially with the TLR4/MD-2 complex, whereas UT41 binds TLR4 both with and without MD-2 association.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse thioglycolate-elicited peritoneal exudate cells. Recommended ≤ 0.5 µg/test (100 µL reaction volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Light Sensitivity: Protect from light (PE-Cy7 is photo-sensitive).
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in dark with minimal signal loss.

Excitation: 488–561 nm
Emission: 775 nm
Laser Compatibility: Blue, Green, Yellow-Green lasers


Target Information

TLR4 (Toll-like receptor 4) is a key component of the innate immune system that recognizes lipopolysaccharides (LPS) from Gram-negative bacteria. It forms a receptor complex with CD14 and LY96 (MD-2) to mediate signal transduction leading to cytokine production.

TLR4 is expressed in placenta, peripheral blood monocytes, B-cells, and myelomonocytic cells. Mutations in TLR4 influence LPS responsiveness. TLR4-deficient mice show increased susceptibility to Gram-negative bacterial infections.
