Target Information
Kaposi’s sarcoma-associated herpesvirus (KSHV) belongs to the gamma-(2)-herpesvirus subfamily and is closely linked to Kaposi’s sarcoma, primary effusion lymphoma (PEL), and multicentric Castleman’s disease.
The KSHV genome (165–170 kb) contains at least 88 open reading frames. Five major proteins are likely involved in HHV-8 capsid assembly: a protease (ORF17), the major capsid protein (ORF25), and three smaller capsid proteins (ORF62, ORF26, ORF65).
Structural studies suggest the KSHV triplex is composed of a monomer of ORF62 and a dimer of ORF26, analogous to HSV-1 and HCMV triplex structures.
