Target Information
Spi-B is a transcription factor belonging to the ETS family, containing a conserved ETS domain that mediates specific DNA binding. It may form homotypic interactions and interacts with IRF4, CREBBP, EP300, SPI1/PU.1, JUN, and TBP. As a sequence-specific transcriptional activator, it binds to the PU-box (5'-GAGGAA-3'), a purine-rich DNA sequence acting as a lymphoid-specific enhancer.
Two isoforms of Spi-B are reported. It is primarily expressed in plasmacytoid dendritic cells (pDCs) and promotes their development while blocking viral replication. Spi-B is not expressed in T-cells or granulocytes but may be present in liver stem cells and B-cells, supporting B-cell receptor signaling, B-cell development, and antigenic stimulation.
