Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IL-21. Anti-Human IL-21-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect hIL-21 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human IL-21 (500-P191) as a capture antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant hIL-21.

Western Blot:
To detect Human IL-21 by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. Used in conjunction with compatible secondary reagents, the detection limit for Recombinant Human IL-21 is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

IL-21 is a 17 kDa immunomodulatory cytokine produced mainly by Natural Killer Cells (NKT), T helper (Th) 17, and T follicular helper (TFH) cells. In TFH cells, IL-21 expression leads to autocrine signaling through the IL-21 receptor (IL-21R) and STAT3, which activates Bcl6 transcription. IL-21 is essential for TFH cell development and function, promotes T cell-dependent B cell differentiation into plasma and memory cells, stimulates IgG production, and induces apoptosis in naive B cells.

In Th17 cells, IL-21 expression and feedback through STAT3, IRF4, and RORγt upregulate IL-23R, facilitating Th17 cell maturation and maintenance by IL-23. IL-21 also downregulates Foxp3. IL-21 deficient mice show protection from chemically induced colitis due to reduced Th17-associated molecule expression, while elevated IL-21 levels are found in colitis models.
