Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IL-21. Anti-Human IL-21-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect hIL-21 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required.
This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human IL-21 (500-P191) as a capture antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant hIL-21.

Western Blot:
To detect Human IL-21 by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL.
Used in conjunction with compatible secondary reagents, the detection limit for Recombinant Human IL-21 is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

IL-21 is a 17 kDa immunomodulatory cytokine produced mainly by Natural Killer (NKT), T helper (Th17), and T follicular helper (TFH) cells.
In TFH cells, IL-21 expression leads to autocrine signaling through IL-21R and STAT3, activating transcription by Bcl6.
IL-21 is critical for TFH cell development and effector function, promoting B cell differentiation into plasma and memory cells, stimulating IgG production, and inducing apoptosis in naive B cells.
In Th17 cells, IL-21 expression regulates IL-23R, IRF4, and RORγt, supporting Th17 maturation and maintenance.
IL-21 also downregulates Foxp3 expression.
IL-21 deficient mice are protected from colitis development, while elevated IL-21 levels are observed in colitis models.
