Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimers or CD8αα homodimers. CD8 binds to MHC class I and associates with p56lck to mediate T cell development and activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes at ≤ 0.5 µg/test (in 100 µL final volume)

Recommended Usage:

• Adjust cell number empirically (10⁵–10⁸ cells/test)
• Titrate antibody for optimal performance
• Add 5 µL of CellBlox Plus Blocking Buffer per sample prior to staining
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains (avoid DNA intercalating dyes such as PI, 7-AAD, DAPI)

Note: NovaFluor Yellow 700 shows non-specific B cell staining in SJL mice, not observed in BALB/c or C57BL/6 strains.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein functioning as a co-receptor with TCR for MHC class I molecules. It is expressed on cytotoxic T cells, NK cells, monocytes, and dendritic cells. CD8 engagement with MHC-I/peptide complexes recruits Lck kinase, leading to T cell activation and cytotoxic response. Differentiation into cytotoxic T lymphocytes (CTLs) is promoted by IL-2, IL-12, and TGF-β1.
