Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule (~32–34 kDa), expressed as a heterodimer (CD8αβ) or homodimer (CD8αα). CD8 binds to MHC class I and associates with p56lck, playing a role in T cell development and activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes (≤0.05 µg/test, 100 µL volume)

NovaFluor dyes compatibility:
Not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample before combining with cells (10³–10⁸ cells/test)

Internal testing shows non-specific B cell staining in SJL mice; not observed in BALB/c or C57BL/6 strains.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor with TCR for MHC-I molecules, playing a key role in T cell activation and differentiation. Expressed on T cells, NK cells, monocytes, and dendritic cells. Activation triggers Lck recruitment, cytokine production, and CTL activation for pathogen and tumor clearance.
