Target Information

CD11a (LFA-1 alpha, ITGAL) together with CD18 constitute leukocyte function-associated antigen 1 (LFA-1), the alphaLbeta2 integrin.
CD11a is implicated in activation of the LFA-1 complex and plays a central role in leukocyte intercellular adhesion through interactions with ICAMs 1–3.
It functions in lymphocyte costimulatory signaling and is expressed on leukocyte plasma membranes in a low-affinity conformation.
Upon stimulation, CD11a shifts to a high-affinity conformation, enabling strong ligand binding.
CD11a participates in immune cell interactions, tissue-specific settlement, and regulation of differentiation, proliferation, and T-cell effector functions.
Blocking CD11a with antibodies or small molecules is a therapeutic approach for inflammatory diseases.
For example, humanized anti-CD11a antibody Efalizumab (Raptiva) is used to inhibit T-cell migration, and simvastatin binding to CD11a leads to immunomodulation.
Two transcript variants encoding different isoforms of CD11a have been identified.
Diseases associated with CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
