Target Information

CD11a (LFA-1 alpha, ITGAL) together with CD18 constitute leukocyte function-associated antigen 1 (LFA-1), the αLβ2 integrin.
CD11a is implicated in activation of the LFA-1 complex and plays a central role in leukocyte intercellular adhesion through interactions with ICAMs 1–3.
It functions in lymphocyte costimulatory signaling and is expressed on the plasma membrane of leukocytes in a low-affinity conformation.
Cell stimulation by chemokines or other signals induces a high-affinity conformation supporting tight binding to ICAMs.
CD11a participates in immune cell interactions, tissue-specific localization, and regulation of T-cell effector functions.
Blocking CD11a with antibodies or small molecules is an important therapeutic approach for inflammatory diseases.
Humanized anti-CD11a antibody Efalizumab (Raptiva) interferes with T-cell migration to inflammation sites, while simvastatin binding to CD11a leads to immunomodulation.
Two transcript variants encoding different isoforms of CD11a have been identified.
Diseases associated with CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
