Target Information
PHLPP1은 serine/threonine phosphatase 계열 단백질로, Akt serine-threonine kinase (AKT1, AKT2, AKT3) 및 PKC (protein kinase C) isoform의 중요한 조절 인자입니다.
PHLPP1과 PHLPP2는 유사한 도메인 구조를 가지며, Akt 활성화에 필요한 두 주요 인산화 부위 중 하나인 Serine473에서 특정 Akt isoform을 탈인산화 및 불활성화시킵니다.
PHLPP2는 AKT1과 AKT3를 탈인산화하는 반면, PHLPP1은 AKT2와 AKT3에 특이적입니다.
PHLPP1은 세포자멸(apoptosis)을 촉진하고 종양 억제 인자로 작용할 수 있습니다. 또한, PHLPP1의 발현 증가가 Akt 매개 인슐린 신호를 방해하여 비만 및 제2형 당뇨병에 관여할 가능성이 있습니다.
