Target Information
Histone H2A.X (H2AX) is a member of the histone H2A family, one of the four core histones forming the nucleosome core particle. In eukaryotes, DNA double-strand breaks (DSBs) trigger phosphorylation of serine 139 at the carboxy terminus of H2AX, resulting in gamma-H2AX. This phosphorylation can be detected by Western blotting or immunofluorescence to assess DSB frequency.
Phosphatidylinositol 3-kinases such as ATM, DNA-PK, and ATR are implicated in H2AX phosphorylation. Structurally, H2A.X contains 143 amino acids. It is synthesized in both G1 and S-phase, with mRNA variants possessing or lacking polyA tails. The gene maps to chromosome 11q23.2–q23.3 in humans.
Histone H2A.X plays a key role in chromatin structure and DNA repair, regulating the interaction of MDC1 (mediator of DNA damage checkpoint protein 1) at sites of DNA damage.
