Bruton tyrosine kinase (BTK) is a cytoplasmic tyrosine kinase belonging to the SRC-related TEC subfamily. Mutations in the BTK gene are associated with severe developmental blocks in B-cell ontogeny and immunodeficiency disorders.
BTK interacts with toll-like receptor (TLR) family members such as TLR4, 6, 8, and 9, which are crucial for innate and adaptive immunity. It also interacts with proteins involved in TLR4 signaling, including MyD88, TIRAP, and IRAK, but not TRAF-6, suggesting BTK’s role in lipopolysaccharide signal transduction.
