Target Information
Oxygen radicals damage chromosomal DNA, causing cell death and inducing mutations. Although much focus has been on repairing 8-oxo-G lesions, many other damaged DNA sites arise from free-radical attacks. The human NTH1 repair protein acts on a wide range of these oxidatively damaged sites.
A homologue of E. coli Endonuclease III, NTH is a DNA glycosylase with apurinic/apyrimidinic lyase activity. The NTH protein exhibits broad substrate specificity, including numerous ring saturation and fragmentation products of pyrimidines.
Research indicates that NTH plays a crucial role in removing oxidative base lesions in mitochondrial DNA.
