Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding allows ER to regulate gene transcription through coactivator or corepressor recruitment.

Two main forms exist — ERα (ESR1) and ERβ (ESR2) — encoded by separate genes. Alternative splicing produces at least four ERα isoforms (66, 53, 47, and 35 kDa). Estrogen receptors are expressed widely, crucial for sexual development, reproductive function, and bone maintenance. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
