Product Specific Information

The HTA125 monoclonal antibody reacts with human Toll-like receptor 4 (TLR4). At least ten members of the Toll family have been identified in humans, characterized by leucine-rich extracellular domains and IL-1 receptor–like cytoplasmic domains. TLR4, along with MD-2 and CD14, forms a complex responsible for LPS recognition and signaling.
TLR4 is expressed by peripheral blood monocytes and can immunoprecipitate human TLR4 (~100 kDa) from transfected cells. Expression levels vary among donors and are generally low on monocytes and other leukocytes.

Recommended protocol: For optimal staining, use the lysed whole blood protocol rather than Ficoll-gradient prepared cells, as density gradient separation reduces staining intensity.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry at 5 µL (2.0 µg) per test (100 µL final volume).
Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

TLR4 is a member of the Toll-like receptor family involved in pathogen recognition and innate immune activation. It is expressed in placenta and myelomonocytic leukocytes. TLR4 forms a lipopolysaccharide (LPS) receptor complex with CD14 and LY96, mediating cytokine production in response to bacterial infection.
Mutations in TLR4 influence LPS responsiveness. Multiple transcript variants exist, though most have uncertain coding potential. TLR4 expression is observed in monocytes, some B cells, and placenta. TLR4-deficient mice show increased susceptibility to Gram-negative sepsis.
