Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells.
It regulates B cell development and maturation by inducing Ig isotype-switching and protects B cells from apoptosis while promoting proliferation.
Interaction of CD40 with CD154 (gp39) on T cells is crucial for T-B cell crosstalk and immune regulation.
5C3 is used for activation of B cells in in vitro functional assays.

Each product contains:

NovaFluor dye compatibility:
Not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Recommended: Invitrogen LIVE/DEAD Fixable Dead Cell stains.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates.
Use 5 µL per sample (10³–10⁸ cells/test).
Add to antibody cocktails before combining with cells.

Target Information

CD40 is a single-chain glycoprotein belonging to the TNF/NGF superfamily. It is expressed on all B cells except plasma cells, and also on epithelial cells, carcinomas, and lymphoid dendritic cells.
It mediates immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
CD40 dysfunction is associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
It may also play a role in Alzheimer’s disease pathogenesis through amyloid-beta-induced microglial activation.
