Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype-switching and, together with IL-4, protects B cells from apoptosis and promotes proliferation.

Interaction of CD40 with CD154 (gp39), its ligand on T cells, is essential for T-B cell crosstalk and plays a role in costimulation and immune regulation. The 5C3 antibody is used for activation of B cells in in vitro functional assays.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis and in vitro functional studies
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.5 µg) per test in a final volume of 100 µL.

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates utilize Phiton™ technology based on engineered nucleic acid dye structures for optimized fluorescent signatures minimizing spillover and spread impacts.

Target Information

CD40 is a single-chain glycoprotein belonging to the TNFR superfamily, homologous to CD30. It is expressed on all B cells except plasma cells, as well as on certain epithelial cells, carcinomas, and lymphoid dendritic cells. CD40 mediates immune and inflammatory responses such as T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

AT-hook transcription factor AKNA regulates CD40 expression, facilitating homotypic cell interactions. TNFR2 acts as an adaptor protein mediating CD40 signal transduction. CD40-CD154 interaction is implicated in amyloid-beta-induced microglial activation, relevant to Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
