Product Specific Information

The HIT2 monoclonal antibody reacts with the human CD38 molecule, an approximately 45 kDa type II transmembrane protein. CD38 is expressed by thymocytes, peripheral B cells including plasma cells, activated T cells, and monocytes. CD38 is a counter-receptor for CD31 and has both cyclase and hydrolase enzymatic activity.

Applications Reported: This HIT2 antibody has been reported for use in flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage is 5 µL (0.125 µg) per test in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®. It emits at 445 nm and is excited with the Violet laser (405 nm). Ensure that your instrument supports detection of this fluorochrome.

Target Information

CD38 (NAD⁺ glycohydrolase) is a type II transmembrane glycoprotein involved in activation, proliferation, and differentiation of lymphocytes, as well as apoptosis of myeloid and lymphoid progenitor cells. It functions as a multi-catalytic ectoenzyme (ADP-ribosyl cyclase, cyclic ADP-ribose hydrolase, NAD⁺ glycohydrolase) or as a cell surface receptor.

Antibodies to CD38 are useful for:

• Subtyping of lymphomas and leukemias
• Detection of plasma cells (e.g., myeloma identification)
• Marker for activated B and T cells

CD38 is expressed in pancreas, liver, kidney, malignant lymphoma, and neuroblastoma. Disorders associated with CD38 dysfunction include chronic lymphocytic leukemia and Richter’s Syndrome.
