Target Information
Glycogen synthase kinase-3 (GSK3)는 glycogen synthase의 조절 인자로 처음 확인된 단백질 키나아제로, glycogen 대사뿐 아니라 단백질 합성, 세포 증식, 분화, 미세소관 조립 및 분해, 세포자멸사 등 다양한 세포 기능 조절에 관여합니다.
GSK3는 두 가지 isoform(GSK3α, GSK3β)을 가지며, 인산화에 의해 엄격히 조절됩니다.
- Ser9 (GSK3β) 또는 Ser21 (GSK3α)의 인산화는 키나아제의 불활성화 유도
- Tyr216 (GSK3β) 또는 Tyr279 (GSK3α)의 인산화는 활성화 유도
활성화된 GSK3β는 p53, c-Myc, c-Jun, HSF-1, cyclin D1 등의 다양한 단백질을 인산화하며, Alzheimer’s disease 진행에 중요한 tau 단백질의 비정상적 인산화에도 관여합니다.
