Target Information

VEGF (vascular endothelial growth factor) is a 45 kDa homodimeric, disulfide-linked glycoprotein involved in angiogenesis, promoting tumor progression and metastasis. It affects vascular endothelium by enhancing cell viability, mitogenesis, chemotaxis, and vascular permeability.

The VEGF family includes VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, and PIGF. VEGF and its receptor system are key regulators of angiogenesis signaling. Most tumors require angiogenesis, with VEGF being a potent angiogenic cytokine. Five isoforms of VEGF are produced by alternative splicing of a single gene, binding primarily to tyrosine kinase receptors FLT1 (VEGFR-1) and FLK1/KDR (VEGFR-2), which are expressed on endothelial cells.

VEGF and its receptors are crucial for embryonic vascular development. A soluble form of FLT1 (sFLT1) binds VEGF with high affinity, inhibiting angiogenesis. Elevated VEGF levels are associated with POEMS syndrome (Polyneuropathy, Organomegaly, Endocrinopathy, Monoclonal gammopathy, Skin changes), also known as Crow-Fukase syndrome.
