Target Information
STAT3는 STAT(signal transducers and activators of transcription) 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동 후 전사 인자로 작용합니다. 사이토카인 및 성장인자에 반응하여 활성화되며, CREB-binding protein 등의 공동활성화 인자가 필요합니다.
STAT3는 Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화될 수 있으며, Tyr705 잔기의 인산화가 이합체 형성, 핵 이동, DNA 결합에 필수적입니다.
인간에서 STAT3 유전자는 17번 염색체 q arm에 위치하며, 다양한 세포 자극에 대한 유전자 발현을 매개하여 세포 성장 및 세포사멸 등 여러 과정에 관여합니다. Rac1은 STAT3의 활성을 조절하며, PIAS3는 STAT3의 특이적 억제 단백질로 알려져 있습니다.
STAT3 유전자의 결손은 생쥐에서 배아 초기 단계에서 치명적이며, 세 가지 상이한 스플라이싱 변이체가 보고되어 있습니다.
