Product Specific Information

This 815CU17 monoclonal antibody reacts with human CD68, a 110 kDa transmembrane glycoprotein belonging to the sialomucin family, related to LAMP and scavenger receptors. CD68 is mainly intracellular, localized in late endosomes, and found on myeloid-derived cells such as monocytes, macrophages, dendritic cells, neutrophils, basophils, mast cells, and certain CD34+ progenitor cells.
It may function in antigen processing or lysosomal membrane protection. Expression has also been observed in activated T cells and subsets of B lymphocytes.

This antibody does not cross-react with mouse or rat CD68.
For flow cytometric analysis of human CD68, clone eBioY1/82A is recommended.

Applications Reported: Suitable for microscopy and immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections.
Applications Tested: Tested for IHC using low or high pH antigen retrieval at ≤10 µg/mL; titration recommended for optimal assay performance.

Filter Recommendation: For eFluor® 570 conjugates, use filters capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Alexa Fluor® 555 or TRITC filters are acceptable.

Target Information

CD68 (Macrosialin) is a 110 kDa integral membrane glycoprotein expressed mainly in lysosomes of monocytes and macrophages, and to a lesser extent in dendritic cells and granulocytes. It may play roles in phagocytosis, lysosomal metabolism, and cell-pathogen interactions. CD68 is expressed in tonsil interdigitating reticulum cells, histiocytic lymphoma, AML, and granulocytic sarcoma. Elevated CD68 expression is observed in CD34+ cells in various malignancies, including acute myeloid leukemia.
