Description:
The BMA12 monoclonal antibody reacts with mouse CD100, also known as Sema4D. Sema4D/CD100 is a 150 kDa transmembrane protein expressed constitutively on T cells and involved in activation of B cells through interaction with CD72. This interaction reduces the negative signaling normally mediated by CD72. CD100 is strongly expressed on resting T cells and weakly on B cells, with expression upregulated upon activation. Antigen expression has been reported in spleen, thymus, and lymph nodes.

Applications Reported:
The BMA12 (BMA-12) antibody has been reported for use in:

• Flow cytometric analysis
• Immunoprecipitation
• Immunohistochemical staining
• Neutralization of mouse CD100 (use Functional Grade purified BMA12, Product #16-1001 for functional assays)

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended usage ≤ 0.25 µg per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody should be titrated for optimal performance.

Purity: Greater than 90%, determined by SDS-PAGE.
Aggregation: Less than 10%, determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Semaphorins are neuronal chemorepellents guiding neurons during development. CD100 (Semaphorin 4D), a 150 kDa homodimer, induces B cell aggregation and enhances viability in vitro. It modulates CD40-CD40L signaling by promoting B cell survival and reducing CD23 expression. CD72 acts as a receptor for CD100, regulating B cell signaling. CD100 is expressed on resting and PHA-stimulated T cells, weakly on NK cells, EBV-transformed B cells, monocytes, and tumor/peripheral blood T cell lines, and at higher levels on activated T and B cells.
