Product Specific Information

The HI111 monoclonal antibody reacts with human CD11a, the 180 kDa integrin alphaL (LFA-1 alpha chain). LFA-1, formed by non-covalent association of CD11a with CD18 (integrin beta2), functions as an adhesion molecule involved in lymphocyte and granulocyte interactions. Its ligands include CD54 (ICAM-1), CD102 (ICAM-2), and CD50 (ICAM-3). CD11a is expressed by all leukocytes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for normal human peripheral blood cells. Use 5 µL (0.6 µg) per test in 100 µL final volume. Cell count range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD11a (LFA-1 alpha, ITGAL) forms part of the LFA-1 complex (αLβ2 integrin) with CD18. It mediates leukocyte adhesion via ICAM-1, ICAM-2, and ICAM-3 and participates in lymphocyte costimulatory signaling. CD11a expression on leukocytes shifts from low- to high-affinity states upon stimulation, enabling tight ligand binding and immune cell interactions. Dysregulation of CD11a is linked to inflammatory diseases such as leukocyte adhesion deficiency. Therapeutic targeting of CD11a (e.g., Efalizumab) demonstrates clinical relevance in immune modulation.
