Background
This locus has a highly complex imprinted expression pattern, producing maternally, paternally, and biallelically expressed transcripts from multiple promoters and 5′ exons. Differentially methylated regions (DMRs) correlate with transcript expression, typical of imprinted genes. Paternally expressed noncoding RNAs may regulate imprinting in this region. One transcript encodes an additional protein, Alex, through an overlapping ORF. Alternative splicing generates various isoforms of the stimulatory G-protein alpha subunit, linking receptor-ligand interactions to adenylyl cyclase activation. Mutations in GNAS2 are associated with several disorders, including pseudohypoparathyroidism types 1a and 1b, Albright hereditary osteodystrophy, McCune-Albright syndrome, and pituitary tumors.
